Growth protocol Mice were sacrificed at 8 weeks. Each sample in the Tg and WT mice are from different individual animals. One mice developed significant scarring and since the array results were significantly different this sample was not used for further analysis. HEK 293 and HK2 cells were obtained from ATCC and cultured as described in Rosenstiel, P.E., et al. Kidney Int 74, 1049-1058 (2008).
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from kidney cortices of five Tg26 and five WT for gene expression microarrays. RNA was isolated by using RNAesay kit from Qiagen.
Label biotin
Label protocol Following standard procedures according to the manufacturer's instructions as applied at the MSSM core facility.
 
Hybridization protocol Hybridization oven 640, rotate 16-18 hrs.
Scan protocol Genechip Scanner 3000-7G.
Description Mouse samples are labeled as investigator, id of sample, condition, gender, date. Human cellline samples are labled as facility, date, investigator, condition, sample id.
Gene expression from kidney coterx from wild type mouse 1
Data processing Expression Console (1.1.2800.29167), affymetrix-algorithm-name = rma
One-way analysis of variance test (ANOVA) with BH correction at 0.05 FDR was applied to the dataset to identify the genes that were differentially expressed between groups.
