Title |
DLD-1/DKK1 cells treated with DOX, DKK1-1 |
Sample type |
RNA |
Source name |
colon cancer cell line
Organism |
Homo sapiens |
Characteristics |
cell line: DLD-1 treatment: colon cancer cells transfected with DKK1
Treatment protocol |
For transduction of DLD1 cells, lentiviruses at a multiplicity of infection of 10-60 were added to the culture medium in the presence of 8 g/ml polybrene (Sigma) for 8 h, typically yielding more than 95% transduced (GFP-positive) cells.
Growth protocol |
All cell lines used in this study were maintained in Dulbecco's modified Eagle's medium containing 10% fetal bovine serum and 2 mM L-glutamine (Invitrogen, Paisley, UK).
Extracted molecule |
total RNA |
Extraction protocol |
Total cellular RNA was isolated using the Rneasy Kit (Qiagen).
Label |
biotin
Label protocol |
Double-stranded cDNA synthesis and in vitro transcription to generated cRNA was carried out using a commercial kit of Affymetrix (Sense Target Labeling kit).
Hybridization protocol |
Samples were hybridized using Affymetrix hybridization kit materials. Heat cocktails at 99° for 5 minutes, then 42° for 5 minutes centrifuge at max speed for 1 minute. Transfer 200μl of hyb solution to each array, then tape holes and parafilm. Hybridize 16 hours at 45° at 60rpm. Fluidics washing is Fluidics Station 450.
Scan protocol |
Affymetrix GeneChip Scanner 7G
