Gene expression profiling of rheumatoid arthritis synovial tissues

(Submitter supplied) Spots were selected that had an expression level higher than 1.6 the local background. The use of a common reference sample allows the comparison of the relative expression levels across the tissue samples. All genes were expressed relative to their median expression level across the arrays of the same microarray platform, allowing us to combine the data from the 2 platforms. Data from Lymphochip and Stanford Human cDNA arrays was combined per patient. more...

Fibroblast-like synoviocytes derived from patients with rheumatoid arthritis

(Submitter supplied) Fibroblast-like synoviocytes (FLS) were cultured from synovial tissues from 19 RA patients. RNA was isolated using the Fast Track 2.0 kit (Invitrogen, Carlsbad, California)from the FLS cultures. Fluorescent cDNA probes were prepared from 1 ug experimental mRNA by oligo dT-primed polymerization using Superscript II reverse transcriptase in the presence of Cy5 labeled dCTP. A common reference mRNA sample (CRE) that consists of a mixture of mRNAs isolated from 11 different cell-lines, was labeled with Cy3. more...

Gene expression profiles in synovial biopsies from patients with arthritis

(Submitter supplied) Rheumatoid arthritis (RA) is an inflammatory joint disorder that results in progressive joint damage when insufficiently treated. In order to prevent joint destruction and functional disability in RA, early diagnosis and initiation of appropriate treatment with Disease-Modifying Antirheumatic Drugs (DMARDs) is needed. However, in daily clinical practice, patients may initially display symptoms of arthritis that do not fulfil the classification criteria for a definite diagnosis of RA, or any other joint disease, a situation called “Undifferentiated Arthritis” (UA). more...

Differential expression of rituximab responders vs. non responders on 3 different blood cell types

(Submitter supplied) New and effective therapeutical options are available for the treatment of Rheumatoid Arthritis. One of such treatments is rituximab, and chimeric anti-CD20 antibody that selectively depletes the CD20+ B cell subpopulation. Similar to established anti-TNF alpha therapies, there is a subgroup of RA patients that do not experience significant clinical response. Therefore, one of the major necessities in actual RA therapeutical management is to identify reliable predictors of the response to this therapies. more...

Rheumatoid arthritis and rheumatoid synovium

(Submitter supplied) Rheumatoid arthritis (RA) is a heterogeneous disease. We used cDNA microarray technology to subclassify RA patients and disclose disease pathways in rheumatoid synovium. Hierarchical clustering of gene expression data identified two main groups of tissues (RA-I and RA-II). A total of 121 genes were significantly higher expressed in the RA-I tissues, whereas 39 genes were overexpressed in the RA-II tissues. more...

Arthritis_RA_OA_VNS

(Submitter supplied) Pigmented villonodular synovitis (PVNS) represents a rare group of lesions with morphological features suggesting an inflammatory as well as a neoplastic nature. Rheumatoid arthritis (RA) is characterized by a chronic inflammation of the synovial tissue and a tumor-like proliferation (TLP) of synovial stroma cells. Osteoarthritis (OA) shows modest inflammatory infiltrates and no TLP. All three diseases result in a progressive destruction of affected joints and remain a diagnostic difficulty because of nonspecific symptoms. more...

Effects of tocilizumab versus methotrexate therapy on gene expression profiles in the early rheumatoid arthrtis synovium

(Submitter supplied) Rheumatoid arthritis (RA) is a chronic, systemic autoimmune inflammatory disease that is characterized by the presence of inflammatory cytokines, including interleukin-6 (IL-6). Here, we investigated the global molecular effects of Tocilizumab, an approved humanized anti-IL6 Receptor antibody, versus Methotrexate therapy, in synovial biopsy samples collected prospectively in early RA before and 12 weeks after administration of the drug. more...

Expression data of subtypes of inflamed synovia from patients with rheumatoid arthritis

(Submitter supplied) Microarrays were used to detail the global program of gene expression underlying differences in the organisation of inflammatory cells classified by the expression of the CD21L and IL-17A genes. Synovia were defined by the expression of the CD21L and IL-17A genes as determined by semi-quantitative PCR.

Identification of novel markers in rheumatoid arthritis through integrated analysis of DNA methylation and microRNA expression

(Submitter supplied) Autoimmune rheumatic diseases are complex disorders, whose etiopathology is attributed to a crosstalk between genetic predisposition and environmental factors. Both variants of autoimmune susceptibility genes and environment are involved in the generation of aberrant epigenetic profiles in a cell-specific manner, which ultimately result in dysregulation of expression. Furthermore, changes in miRNA expression profiles also cause gene dysregulation associated with aberrant phenotypes. more...

Expression data from active rheumatoid arthritis patients and healthy control.

(Submitter supplied) Rheumatoid arthritis (RA) is a systemic autoimmune disease and its underlying molecular mechanisms are still poorly understood. Previously a CD4 T-cell microarray study has only focused arthritis patients. We aimed to compare the molecular profiles of active RA versus healthy control in CD4 T cells. We used microarrays to detail the global programme of gene expression in active RA and healthy control.

RHEUMATOID ARTHRITIS SYNOVIAL MICROENVIRONMENT INDUCES METABOLIC AND FUNCTIONAL ADAPTATIONS IN DENDRITIC CELLS

(Submitter supplied) Rheumatoid Arthritis (RA) is a chronic autoimmune disease which causes degradation of cartilage and bone. It is well appreciated that the pathogenic hallmark of RA is the mass influx of inflammatory cells into the joint. However, the role dendritic cells (DC) may play in this inflammatory milieu is still relatively unexplored. Moreover, the contribution this unique synovial microenvironment has on DC function and maturation is still unknown. more...
