Overall design |
The experiment was performed as described previously with modifications (Kumar et al., 2019). Briefly, Group 3 medulloblastoma PDXs (Med-411FH, Med-211FH, BT2019051) were engrafted into NSG mice. Once the animal reaches humane endpoint, Hoechst 33342 (2.5mg/kg body weight) was injected through the tail vein and after 15 min the tumor was resected. Tumors cells were dissociated using enzyme-free cell dissociation solution (Millipore # S014C) and the single cell suspension was passed through a 70 micron cell strainer. The tumor cells are sorted based on Hoechst uptake using BD FACSAria III cell sorter.
