Mice were exposed continuosly to a diet containing 700mg/Kg of Doxocyclin (Gordon’s Specialty Stockfeeds) from week 6 of age until the ethical endpoint (8 weeks total treatment)

MMTV-PyMT mouse mammary tumor samples (and metastasis) were processed for single cell suspension and FACS sorted based on Lin- CD24+ markers. RNA was extracted from these populations straight after FACS sorting.

Extraction protocol |
Total RNA was extracted by using the Qiagen RNeasy minikit (Qiagen, Doncaster, Vic, Australia) according to manufacturer’s protocol. Following extraction, RNA concentration and purity were analyzed using the NanoDrop® ND-1000 Spectrophotometer (Nano-Drop Technologies, Wilmington, DE, USA). For Affymetrix GeneChip® analysis, the integrity of RNA was analyzed using the Agilent Bioanalyser 2100 6000 Nano Assay (Agilent Technologies, Palo Alto, CA, USA) according to the manufacturer’s instructions.
