Treatment protocol |
Treated animals were injected with a single 12.5mg/kg dose of Pip6e-PMO in sterile saline via the tail vein at 12 weeks of age.

Growth protocol |
Tibialis anterior (TA) muscles were macrodissected and snap frozen in liquid nitrogen-cooled isopentane. 8mm cryosections were prepared from mid-belly of the muscle.

Extraction protocol |
Samples were extracted using the TRIzol method and DNase I treated with Ambion TURBO DNA-free according to manufacturer's instructions. Samples were re-extracted using acid phenol chloroform and ethanol precipitated. Each RNA sample was analysed using the Agilent 2100 Bioanalyzer (Agilent Technologies, USA) to assess RNA quality.

500 ng of total RNA was labeled using the FlashTag Biotin HSR kit (Genisphere).

Total labeled RNA was hybridised to the Affymetrix Genechip miRNA array 3.0 using manufacturer's protocols.

Image files from scanning (.dat and .cel files) were generated with Affymetrix Expression Console package.

Data processing |
The processing and data analyses were performed in Affymetrix Expression Console Software. The arrays were analyzed using Robust Multichip Analysis (RMA) and DABG (detected above background). In the final analysis we sorted out Mus musculus transcripts only. Transcripts with Absent detection signals in at least three of the four replicates (or two out of the three replicates in the controls) were removed.
