Extraction protocol |
Total RNA was extracted and isolated using mirVana TM RNA Isolation Kit (Applied Biosystem p/n AM1556) following the manufacture’s introduction. The concentration of RNA was measured by NanoDrop spectrophotometer. Total RNA concentration ranged form 100ng/ul to 1ug/ul. The RNA quality was evaluated by Agilent 2100 Bioanalyzer; RNAs with RIN>=7.0 and 28S/18S>0.7 were accepted for microarray analysis.

100ng of total RNA was reverse transcribed into cDNA and labeled with Cy3, and then hybridized on a miRNA array (Agilent-021827 Human miRNA Microarray (V3)) using Agilent’s miRNA Complete Labeling and Hyb Kit (p/n 5190-0456).
