Androgen sensitive and insensitive prostate cancer cell lines: expression profiles

Analysis of androgen sensitive (AS) and insensitive (AI) prostate cancer cell lines. Despite the wide use of these cell lines as model systems, a global genotypic characterization of these cell lines is currently lacking. Results identify differences in gene expression profiles.

Genome-wide characterization of gene expression variations and DNA copy number changes in prostate cancer cell lines

(Submitter supplied) Abstract: BACKGROUND: The aim of this study was to characterize gene expression and DNA copy number profiles in androgen sensitive (AS) and androgen insensitive (AI) prostate cancer cell lines on a genome-wide scale. METHODS: Gene expression profiles and DNA copy number changes were examined using DNA microarrays in eight commonly used prostate cancer cell lines. Chromosomal regions with DNA copy number changes were identified using cluster along chromosome (CLAC). more...

Combined arrayCGH and SNP-loss of heterozygosity analysis in cervical cancer

(Submitter supplied) BACKGROUND: Cervical carcinoma develops as a result of multiple genetic alterations. Different studies investigated genomic alterations in cervical cancer mainly by means of metaphase comparative genomic hybridization (mCGH) and microsatellite marker analysis for the detection of loss of heterozygosity (LOH). Currently, high throughput methods such as array comparative genomic hybridization (array CGH), single nucleotide polymorphism array (SNP array) and gene expression arrays are available to study genome-wide alterations. more...

Integrative Genome-wide Analysis of Glioblastoma.

(Submitter supplied) Glioblastoma multiforme shows multiple chromosomal aberrations, the impact of which on gene expression remains unclear. To investigate this relationship and to identify putative initiating genomic events, we integrated a paired copy number and gene expression survey in glioblastoma using whole human genome arrays. Loci of recurrent copy number alterations were combined with gene expression profiles obtained on the same tumor samples. more...

Genome-wide screening for complete genetic loss in PCa by CGH onto cDNA arrays

(Submitter supplied) We demonstrate that comparative genomic hybridisation (CGH) onto cDNA microarrays may be used to carry out genome-wide screens for regions of genetic loss, including homozygous (complete) deletions that may represent the possible location of tumour suppressor genes in human cancer. Screening of the prostate cancer cell lines LNCaP, PC3 and DU145 allowed the mapping of specific regions where genome copy number appeared altered and led to the identification of two novel regions of complete loss at 17q21.31 (500 kb spanning STAT3) and at 10q23.1 (50-350 kb spanning SFTPA2) in the PC3 cell line. more...

High-resolution gene copy number and expression profiling of human chromosome 22 in ovarian carcinomas

(Submitter supplied) This set contains the aCGH data for the publication. Previous low-resolution studies of chromosome 22 in ovarian carcinoma have suggested its involvement in the development of the disease. We report a high-resolution analysis of DNA copy number and gene expression of 22q in 18 ovarian carcinomas using a 22q-specific genomic microarray. We identified aberrations in 67% of the studied tumors, which displayed 3 distinct gene copy number profiles. more...

Intratumoral heterogeneity of prostate cancers

(Submitter supplied) Copy number analyses of regionally separated intratumoral biopsies of prostate cancers. Intratumoral heterogeneity (ITH) leads to regional biases of the mutational landscape in a single tumor and may influence the single biopsy-based clinical diagnosis and treatment decision. To evaluate the extent of ITH in unifocal prostate cancers (PCAs) that had not been sought, we analyzed multiple regional biopsies from three PCAs using DNA copy number analyses. more...

Taxane-selected ovarian cancer cell lines

(Submitter supplied) Each of the six ovarian carcinoma cell lines was exposed to docetaxel or paclitaxel at IC50 levels (the concentration required to kill 50% of the population). After several passages at this initial concentration of taxanes, drug concentrations were escalated, and this process was repeated until variants displayed at least a 10-fold resistance to the selecting agents. mRNA and genomic DNA were isolated from the parental and resistant variants, and hybridized onto Stanford human cDNA microarrays containing about 42000 elements. more...
