Whole-genome microarray analysis of human skin fibroblasts

(Submitter supplied) Connective tissue growthfactor (CTGF/CCN2) is a matricellular protein induced by transforming growth factor-beta (TGF-beta) and intimately involved with tissue repair and overexpressed in various fibrotic conditions. We have previously shown that keratinocytes in vitro downregulate TGF-beta induced expression of CTGF in fibroblasts by an interleukin-1 alfa (IL-1alfa) dependent mechanism. With this study we want to get a better overall perspective who fibroblasts respond on TGF-beta and in a TGF-Beta stinulated system, what effect addition of IL-1 alfa have. more...

miRNA expression data in unstimulated and TGF-β1-stimulated lung fibroblasts

(Submitter supplied) Lung fibroblasts play an important role in extracellular matrix homeostasis and this process is mainly regulated by transforming growth factor-beta (TGF-β). Hence, lung fibroblasts are postulated to play a crucial role in aberrant lung tissue repair and remodeling, which is a main factor in lung diseases like COPD. In this study, the effect of TGF-β1 on the miRNA expression in parenchymal lung fibroblasts is investigated.

Expression data from human primary fibroblasts treated with Trypanosoma cruzi-conditioned medium

(Submitter supplied) The intracellular pathogen Trypanosoma cruzi secretes an activity that blocks TGF-β-dependent induction of connective tissue growth factor (CTGF/CCN2). Here, we address the mechanistic basis for T. cruzi-mediated interference of CTGF/CCN2 expression by examining host cell signaling pathways and the global inhibitory effect on TGF-β-dependent gene expression. We show that the expression of a discrete subset of TGF-β-inducible genes involved in cell proliferation, wound repair, and immune regulation are blocked by the soluble T. more...

Transforming growth factor-β effect on fibroblasts in the presence of Trypanosoma cruzi conditioned medium

Analysis of fibroblasts stimulated with TGF-β in the presence of T. cruzi-conditioned medium (PCM). A soluble factor(s) from T. cruzi inhibits TGF-ß-induced MAP kinase activation. Results provide insight into the ability of secreted parasite molecules to hinder agonist-induced host cell signaling.

TGF-beta regulated gene transcription and protein expression in human GFAP-negative lamina cribrosa cells

(Submitter supplied) Primary open angle glaucoma (POAG) is a progressive optic neuropathy, which is a major cause of worldwide visual impairment and blindness. Pathological hallmarks of the glaucomatous optic nerve head include retinal ganglion cell axon loss and extracellular matrix (ECM) remodeling of the lamina cribrosa layer. TGF-beta is an important pro-fibrotic modulator of ECM metabolism, whose levels are elevated in human POAG lamina cribrosa tissue compared with non-glaucomatous controls. more...
