RNA |
Source name |
Mouse liver, normal diet, 16 weeks
Organism |
Mus musculus |
Characteristics |
tissue: liver treatment: normal diet strain: C57/BL6 gender: male age: 8 to 10 week old
Growth protocol |
Liver tissues were collected and frozen immediately in liquid nitrogen.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed using mirVanaTM RNA Isolation Kit according to the manufacturer's instructions.
Label |
Biotin
Label protocol |
Total RNA were tailed with Poly A and then labeled with Biotin.
Hybridization protocol |
The labeled RNA were hybridized for 16 hr at 48°C on Affemetrix miRNA 3.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.
Scan protocol |
The arrays were scanned by the Affymetrix Scanner 3000 (Affymetrix).
Description |
non-exercised mice remained sedenrary
Data processing |
Affymetrix GeneChip Command Console software (version4.0, Affymetrix) was used to analyze array images to get raw data and then offered RMA normalization. Next, Genespring software (version 12.5; Agilent Technologies) was used to proceed the following data analysis. Probes that at least 75 percent of samples in any 1 condition out of 2 conditions have flags in P were chosen for further data analysis. Differentially expressed miRNAs were then identified through fold change as well as P value calculated using t-test. The threshold set for up- and down-regulated genes was a fold change>= 2.0 and a P value<= 0.05.
